Yes, the Apple TV box has a ethernet plug just for that.

Actually, you should use a wireless router to connect to your Apple TV. It eliminates having to run any extra cables behind your entertainment center! If you buy an N-Series router, you'll be able to push large files even faster. In fact, that is one of the best features of the Apple TV unit anyhow... =)
