I'm assuming you are not talking about the blank rune "Wyrd" but rather one of the other runes lying face down... I simply turn it over and read it as such. This is effective if you are not casting ALL of the runes. If you are throwing all of them onto a cloth, then read the ones that are face up. (SRW suggests this in one of her books... I never did like that method) I draw mine one at a time from a special pouch I made.
Hope that helps.

If the Blank Rune appears in your yes or no draw, it means one of two things: either the question is not ready or able to be judged, or you are better off not knowing the answer. After waiting a day, you can ask the question again. If the Blank Rune appears again, you can try asking the runes a third time. However, if the Blank Rune appears a third time, then you really should give up asking the question because you are simply not meant to have an answer.

The best thing to do is toss out the "blank" Rune. Throw it in the bin. There is ZERO historical basis for it, and it was, in fact, tossed in the mix so there would be an even 5 sets of 5 Runes. It's a purely modern contrivance.
Toss it and understand that the Runes consist of 3 Aettir of 8 Runes each. They belong in those Aettir for a reason.
So... first things first, toss the blank, or use a dremel and make a talisman out of it.

I agree with the previous answers about discarding the Blank Rune, but that's not the way I read your question, so I'm going to answer what I understand you to be asking.
If the stone is face-down, you can approach it one of several ways.
Some scholars/authors will advise that if the stone is face-down, it should be treated as "murk-stave" - usually an opposite meaning to the rune's 'normal' meaning.
Others advise to leave it as it is and draw another to take it's place.
Personally, I just flip it over and continue with the interpretation. The stone and the message are already there - but like all things related to knowledge, sometimes it takes a little effort to get the answers.

There is alot of information on the meanings of the runes on the web. Many rune casters don't even include the blank rune, it's a modern invention...
