Certainly with confidence. A prospective boss wants to know you are willing to do your job efficiently and in the best way possible. They want to hear your desire to do well and want to know you are sincerely interested in the position. Make your answer relavent to the job. If it is as a receptionist for example, tell them you have the best telephone skills; you take pride in your job; you believe in always smiling over the phone and being very understanding of all the clients who walk into the door. Confidence is key. If you don't have it, you wont win the job.
