NaOH = to provide the alkaline solution.
CuSO4 = so that the copper (II) ions and peptide bonds react within the alkaline solution.

The biuret test is a reaction between copper ions and peptide bonds in an alkaline solution.

NaOH is a weak alkali, thus it is suitable 4 the test. CuSo4 bcos it reacts w/ the peptide bonds in the solution. Other than that, the only reason i can think of is that they're both in the Biuret reagent.
