Breathing pure helium without oxygen can damage brain cells due to lack of oxygen. Helium should not damage your voice box. The difference in density relative to air alters the vibrations of the voice, especially pitch. Heliox is a mixture of helium and oxygen used for very deep diving to avoid nitrogen narcosis.
