Federal laws (Americans with Disabilities Act) covers this. I don't know if Florida has specific laws, but businesses must comply with the federal laws. Most motels and hotels have at least one, and larger facilities have several, wheelchair accessible rooms.. You will want to call ahead and make reservations, as the room/s could already be booked; or, after a certain time, if a handicapped room is not rented, and the hotel is filling up, they will rent it out to non-handicapped.
The ADA laws apply to any place of lodging having 5 or more rooms. In my experience, unfortunately, most B & B's are not accessible, nor required to be under federal laws.
