There are some variables for picking the "best". As far as materials, I like Lowes and Home Depot. The problem is the sales people who are not always as knowledgeable as advertised on TV. Ace Hardware seems to always have someone with a good amount of expertise.

TOOLS - NOT Harbor Freight or WalMart - they are the cheapest foreign made crap there is. Even the national brand names admit to making their stuff cheaper for WalMart.
A local owned and operated place is always the best. Contractor supply houses are the best for product knowledge, tools and advice. The national chains have good prices, but also have minimum wage clerks who shouldn't be allowed to tell you how to boil water.
If you don't have a contractor supply house, then go to a locally owned hardware store.

It depends on what your looking for:
For tools no one beats harbor freight
http://www.harborfreight.com/
For hardware "Ace" is the place.
For DIY electronics components
http://www.octopart.com/ (part search - good for replacing broken parts)
http://www.sparkfun.com/
http://www.mpja.com/
For materials and kits (i.e. tubs, sinks, flooring, shelving) goto the big box stores.
Homedepot
Lowes
Walmart (their selection is limited but what they have is very cheep)
