Well, many times when I didn't have money for a stump grinder, I dug a bigger hole around the the roots, then I just started to hack with a hatchet, and saw when I could, then dig out a little more earth, repeat hacking and sawing etc.
Finally when I had all but the tap root I would use the tractor to pull the remainder of the stump out.
You did say Manual Labor! Really there are easier ways how ever.

Oh By the Way if you use the Rotting liquid, do not plan on re-planting for a few years, I have found that NOTHING will grow on that spot for 5 years or better!

First off, just how big is your large tree stump? Get a tape measure and measure the width and height.
If its ground level, your in big trouble. Not that it will be impossible, but Id never try it. At least 4 ft of height is benificial for leverage.
Depending too on how 'large' it is, you might be able to hire it done relatively cheap. Some stump grinding guys charge by the inch. If youre willing to clean up the mess, it can lower the cost.
As for your original question: start 2-3 ft out from the edge, start digging downward cleaning as much dirt from the hole as possible. when you hit a root, chop it with a SHARP axe on both ends (take out a 2-3' long peice-this will allow you to continue to dig w/o it getting in the way). As you get deeper, start digging under the stump, chopping roots as you go along. If you have some height left, as you get more roots cut and a deeper hole, you should start to get it loose. You may have to put on some sort of mechanical advantage (truck, tractor, etc.) DONT PULL TO HARD! If you break your pulling arm (rope/chain) it can cause damage to your vehicle.

If you are in a hurry, follow the advice about a stump grinder, if it isn't too large, you can chain it to a tractor or truck and try to drag it out. Neither one is "manuel". If you have a few years, you can hack out a depression in the center for water to pool. that will hasten rot, and give you a bird bath at the same time.

Good luck, it is a lot of hard, back breaking work. Unless, you have a back hoe and then you can get roots and all. Otherwise, an ax, chain saw and strong arms will do it.
There is a product called stump remover that is an acid base and it will kinda melt the center and make it a little easier. Good luck.
