Heart disease. His father, in fact, never saw Darryl pitch in the majors, as he died very young as well. Sometimes early cardiovascular disease is both hereditary and fatal.
It was June 22, 2002. His last start was 4 days earlier, just hours before Jack Buck passed away. In that game, Kile pitched the Cardinals into first place in the division, and they never relinquished that spot for the rest of the season. Joe Girardi, who hwas the Cubs regular catcher (this was in Chicago), made the announcement to the fans that the game was "cancelled due to a tragedy in the Cardinal's family", although at the time of that announcement, Kile's wife hadn't yet been notified, so the nature of the tragedy wasn't announced.
It was a Saturday, and that game was supposed to be Fox's A game. Joe Buck was to do the play-by-play, in his first broadcast since his father's passing. Tony LaRussa did his best job of managing in that season. He kept that team going, and did it despite the fact that not long before Kile's untimely demise, LaRussa had lost his mother.
All around, a tragic day and period for baseball in general and the Cardinals' organization in particular.
Kile had also pitched for the Astros and Rockies., and had a no-hitter with the Astros.

Darryl Kile was a pitcher for the St. Louis Cardinals. He died in his hotel room in Chicago; the Cardinals were there to play the Cubs.
He died from some type of heart ailment, which was never discovered prior to his death.

On June 22, during pregame warmups for what would have been a day game against the Cubs, team personnel noted Kile's absence. Hotel staff entered Kile's room and discovered him in his bed, under the covers, dead of a heart attack at age 33.

He died of a heart attack that he suffered in his hotel room in Chicago when he was to play the Cubs. Nobody knew until he didn't show up to the game.

It was a heart attack. He had some sort of rare genetic problem with his heart, that wasn't discovered until after he died, that led to the coronary.
