I think the best sketch they've ever done is the Humour lecture (aka the custard pie sketch or the jape sketch) they performed in many of the live shows (incl. Live at the Hollywood Bowl). If you don't know it, it involves Graham Chapman delivering (in a very deadpan way) thebest ways of tripping people up, wopping them on the head with planks and delivering a custard pie, whilst Michael Palin, Terry Gilliam and Terry Jones demonstrate.

Although it was written by Terry Jones and Michael Palin pre-Python (actually whilst they were still at Oxford University), it has got to be the pinacle of slapstick writing.

There's a recording on YouTube (link below) if you want to watch it and judge for yourselves (the film isn't the best quality but you'll get the idea).
