The best way to seal a deal in a job interview is to be confident and prepared.

Don't sell yourself short either, most employers and HR people are skilled at knowing whether or not you have the raw talent to do the job even if you don't have the exact skills they are looking for.
When an employer takes on a new employee, they are taking a chance and making a financial investment. Make sure that if you really want the job that you present yourself as a worthwhile long term investment.
