Yes, you can convert FAT16/32 to NTFS without losing data. Use Paragon's partition manager for this. Great and (that is much more important) stable utility, I use it for a long time. Program's website:

PS There're also Symantec's partition magic, but it has no vista compatibility and most likely there'll no updates and upgrades at all for this tool, that means poor hardware compatibility and no new features. Another tool - Acronis disk director is VERY unstable, it crashed my hard drive during spliting partition :((( and I moved to paragon. Actually, I don't know if they can convert file systems or not.

You can change file system without any additional software. All you need is to use "convert" command it command prompt (start>>run>> type "cmd"). As for Disk Director unstabilty I think that the person above just can't read manuals carefully or have some problems with hands, this opinion is based on the personal experiece since I've been using this program for a long time and it works like a charm.
