You want to start pretty early. I'd say 8am, and continue your sale until:
a. your run out of things to sell
or
b. you get bored.

Most garage sales are held on Friday and Saturday (some extend into Sunday) because thats when people are out shopping for bargains. Start early in the morning. Eight is a good time, but if you are advertising it, state that there are no early sales. (you will still have people there early) Be ready when your posted starting time arrives. Hang clothes. You will have a disorganized mess by 9 if you just fold and lay them out. We use masking tape with the price and initials of the seller (if there is more than just one family or individual). You need to pay attention for tag swappers. Prices for a garage sale should be "bargain prices". Most things are sold for 1/2 or less than retail. Unless you want to take the leftovers back in the house at the end of the day. We always have a board to stick the tape with prices onto so at the end of the day you have a "list" of who sold what.

People expect a garage sale on a Saturday morning. Fanatics will be out by 6AM hunting down bargains, so if you advertise 8, be up at 5:30 to haul your junk out. Some people will also do a Friday afternoon or a Sunday AM, but Saturday from about 6 to noon is where you're going to make money.
As far as pricing, you can put "5 cents" and some jerk is going to haggle you down to 3 cents. That's just how the game is played, so expect it.
Organization- get some card tables. When I drive by a sale and everythign is just on the ground on a blanket, I figure its just junk & i might not stop. Also have a couple of big items to lure people in. The more you have out, the more interest you'll get.
And lastly, put signs with arrows up all over the place! And put the date, too, so that when you forget to take the signs down by next weekend, people aren't looking for nothing. But don't put your address or the cops in a really strict neighborhood will know who to fine for littering.

Do it on a sunny Saturday, because that's when people are out. Also Friday afternoons are good. When you put everything out, try and put the similar stuff together, books with books, clothes with clothes, etc. If you can get more specific (baby clothes with the other baby clothes, near the maternity clothes) it's better, but not necessary. As for prices? Price to sell. People don't go to yard sales for $10 shirts - they go for $1 shirts. So try and keep prices low, low, low. Furniture can be higher priced, obviously. Lastly, be willing to negotiate. Bargaining is a staple of yard sales.

personally i like to have a garage or go to a garage sale early in the morning say start it from like 7:30 to 5:30. This way you will give people the chance to have time to come buy. The prices should be reasonable or you wont be able to sale a thing. say like clothing tshirts should go like 50 cents shoes should go 1dollar. jeans should go like 1dollar 50 cents. toys 50 cents to 2 dollars. i think you should keep your prices low and reasonable if you do this you will sell more and make more money. If you have kids have them set up a lemonaide stand and sell lemonade for like 50 cents a cup its a good way for them to make money also and have fun and (stay out of your hair.)
