Depends on the machine. You need to read up on your user's guide. Typically the larger Ram goes into the 1st slot on older machines. Some motherboards require that both slots contain the same amount of memory, like 256 or 500 for each. And some machines can not handle more than x memory.
Seriously, you need to take your computer's name and model number and do some research at the manufacturer's site to make sure that you don't mess up your motherboard and the new ram.
But if the machine is less than 12 months old you might be safe in just putting the larger ram into the 1st slot.

YES, well if it fits in the slot then it doesn't matter but it has got to be the same type of ram like the same company makes it other wise i guess it doesnt matter and the different sizes shouldnt make a diff i do it all the time ive made my own computer like that

Assuming that:
1) Both RAM sticks are compatible with the computer, and
2) Your computer is capable of handling 2 GB of RAM
Then it doesn't matter which RAM goes in which slot.
If your computer can only take one gig of RAM, then you'll need to take the gig stick back and buy two 512s. Then, obviously, you'll have to remove the already-installed 256 stick, put a 512 in each slot, and then your computer will have a gig.

closest to the processor but you should match your ram if you have a 512 stick you should only add a 512 stick never mismatch two different size ram modules

at the start, get the memory, the pc and a huge hammer... placed the memory on perfect of the pc and use the hammer to pound it into the pc. If that don't artwork, you will would desire to get a clean pc. i'm purely kidding. do not try this.
