If we just stick within the Marvel realm, its debatable. There are tons of characters who are much stronger, much more powerful, invulnerable, etc... but Spidey's strength has always been his intelligence and his clever use of all his abilities. Whereas someone like Thor could technically squash him like a bug (no pun inteneded), Spidey is fast enough to keep him on his toes, while utilizing webbing and banter (yes that is a power and Spidey is the master) and Spidey sense towards one main goal; keep Thor off balance. You don't want to get struck even once by someone like Thor, so Spidey's tactics are to keep his adversaries off balance and guessing all the time. In the meantime, Peter uses his genius to think his way out of any spot.
Of course he's not undefeatable. Thor may eventually wear him down with sheer might. He's very mighty. Spidey really couldn't do much against the mega characters of Marvel like Dr. Strange, Thanos, Silver Surfer, etc.
More along Spidey's weight class, i think it would come down to tactics. Spidey's been around for a long time, and has had a long time to become an instictive fighter. That combined with the intelligence factor makes him difficult to beat. To do that, one would have to be a master combat tactician and intensely focused, not easily being drawn off balance by Spidey's wiley banter & speed. Captain America would be the obvious choice, being the ultimate tactitian and hand to hand combatant (until he died, i wonder if that was his strategy?). Venom is a near equal, and has intimate knowledge of his way of thinking. Wolverine, through sheer tenacity. Daredevil? Proffessor X (He's faking. He can walk).
