Depending on the make and model; most weed eater spools come out by simply pulling straight out on the spool itself. If this does not work, push down on the spool and slide in either a clockwise or counter-clockwise direction (depending on which way it slides) and begin applying a pulling force. Use caution in doing so - as to not lose the spring that assists in letting the line out.
Once out - wind the line in the direction of the arrow and bend the line into one of the groves made to hold the line (this help you replace the spool). Once everything is back together - pull on the line slightly to release the line from the groove and you are ready to go.

Is there a nut securing the bottom? If not, most are a press down and rotate counterclockwise on the bottom while holding the rest in place. The bottom cover most likely has a couple tabs that twist lock.

They screw off, but it might be on tight so you will have to use some muscle. (remember: righty tighty, lefty loosey)
Note: there will be a spring under the spool...don't lose it!
