When a computer is powered on, data from the hard drive is loaded into RAM so the processor can execute the code in memory.
The BIOS code in ROM is ran when first powered on to tell the processor to fetch the data from the Hard drive.

Instructions in a computer program read data from the disk into an area of RAM used by the program. Windows takes over to handle the data transfer. Normally, the instructions will be passed to the disk controller and Windows or a program can run other things whilst the controller transfers the data to RAM. That is known as Direct Memory Access (DMA) mode.
