First of all its imp to know what type of protein the blood ur tyring to estimate,based on which either serum or plasma is used for analysis.serum is without blood factors(which again are proteins)otherwise it contains all other proteins.

Blood plasma contains many vital proteins including fibrinogen, globulins and human serum albumin. Sometimes blood plasma may contain viral impurities which must be extracted through viral processing.

"Serum" refers to blood plasma in which clotting factors (such as fibrin) have been removed naturally by allowing the blood to clot prior to isolating the liquid component.

Well the biuret method depends on the shade of the sample after testing to indicate protein presence therefore i'd assume that because serum and plasma are made up of different constituents there is different quantities of peptide bonds and so different amounts of proteins so producing a different shade according to the amount.
