Just got my pink DS. Ended up buying Wario: Master of Disguise to start out with since they did not have any of the others that I wanted and it was only $25 from the sale Target is running on games.
Keep the answers coming though, I'd like to get more suggestions. :)

When it started, DS didn't really have a good selection, but now it has some great games.
If you like old-school side-scrolling Mario, get New Super Mario Bros. It's like Super Mario World (SNES) but Nintendo revamped the graphics, making all the characters 3D, and gave Mario some new items.
If you like racing, get MarioKart. Nothing beats the rush of being in second place thirty feet before the finish line, pummeling the leader with a red turtleshell and zooming past for the win.
If killing zombies and leveling up are your thing, try Final Fantasy III. It's a slow, but rewarding RPG with a deep storyline and lots of twists and turns. Also, for a more "action-packed" zombie-killing rampage, go with Castlevania: Dawn of Sorrow, or Castlevania: Portrait of Ruin. Both are excellent, excellent games with great and well, good storylines, respectively, and they both have incredible graphics and music for handheld games.
Lastly, if you enjoy strategy games, you can take home Advance Wars: Dual Strike. This turn-based strategy game pits your army against another as you build your army, capture cities, deploy fighters, launch cruise missles and destroy your enemies. This game is fairly long with 29 missions, a Hard mode, more than 80 maps for points, and literally hundreds of unlockables. Alternate uniforms for your commanders, music mode, gallery mode, combat mode.... Advance Wars never disappoints.
Anyway, hope this gives you some ideas. Sorry it's so long.

Brain Age and Big Brain Academy are fun...they actually require some thought, but at the same time, you can finish a segment quickly. The New Super Mario Bros. is really good, as is Tetris DS...because who doesn't like Tetris? It's a classic!
My boyfriend loves Final Fantasy III. My brother said that Hotel Dusk was boring.
I just got my DS not too long ago, so I am interested in the answers too! :)

i recently bought an NDS about 3 weeks ago and have been researching the subject extensively. here are some top games to name a few:
mario kart (racing)
kirby's canvas curse (platformer)
new super mario bros. (platformer)
yoshi's island (platformer)
castlevania - portrait of ruin (platformer)
Some other NDS games that I've played and enjoyed:
Mario vs. Donkey Kong 2: March of the Minis (mini games)
brain age (puzzle)
lost in blue (rpg)
mario touched! (mini games)
Phoenix Wright: Ace Attorney (storybook)
Elite Beat Agents (rhythm)
Trauma Center: Under the Knife (simulation)
i was dissapointed with the limited selection of double-screened playable platformers for the NDS. the only two available are Sonic Rush and Yoshi's Island DS. hopefully there will be more in the future. In the meantime you can play gameboy advanced games using the slot-2 input. one of my favorite GBA platformers is Drill Dozer -- this is mindless platform fun that uses all of the buttons to do cool moves. Donkey Kong country 3 is also really good.
most games can be purchased used from amazon or half.com for a significant discount. online is a great resource for titles that are no longer made. stores like target are only going to stock the latest and greatest titles, and most of the time the shelves are empty.
