What can't you do?
This is what I would do:
- day trip to Victoria (the ferry ride's really nice. In Victoria you can go to the Royal BC museum amongst lots of other things)
- Chinatown
- Gastown
- Granville Island
- Museum of Anthropology, followed by a walk down to Wreck Beach (at UBC)
- Stanley Park (you can easily spend at least 1/2 day there)
- go for a walk up a the top of Grouse Mt. or Mt. Seymour
- go up to the park on top of Burnaby Mt., near SFU - nice view of the city.
- drink some local beer
- eat some West Coast food (unless you're from the West Coast already and have had it all: smoothies, wraps, BC rolls, etc.)
- after that it depends on your personal interests. There's lots to do in Vancouver, however if you're really into clubbing and nightlife, it's sort of a quiet city. It exists, but not to the extent of other big cities.
