If the 'wanna' means 'wanted to' the right phrase would be

Wanted to speak but couldn't

If the 'wanna' means 'want to' the right phrase would be

wanna speak but can't

"I would have liked to have spoken but I could not" is what you are looking for. And there is no such word as "wanna"; it should be "I wanted to speak......" etcetera.

Even your second question is not correct or well written. It should be "Is this the correct what to use the word couldn't?"

"I wanna(=want to) speak but can't" is correct because present tenses are used.
"I wanted to speak but couldn't" is also correct because past tenses are used.
