How are they different?

First off, there is no "e" in the word grammar.

I presume the difference in #2 is a space between the comma and the quote - that is correct. Be sure your punctuation mark goes inside the quote.

I came into the room and said, "Hello."

Do you mean the line jump?
You should always start the beginning of the quote (the "he said" part) in a new paragraph, not just the quote.
(new paragraph or line indention here) He said, "This way is right!"
Not like this:
He said,
(indention here) "This is wrong!"

#2 is correct. There should be a space after the comma.
And hey, no "e" in grammar- it's an "a".
