To answer your question: Day #1 would be the first day of your period.
But -- and this is a significant "but" -- that generalization is based on the notion that a woman has a 28 day cycle and ovulates on day 14 of that cycle. The fact is, that generalization does not apply to many, if not most, women. (And even if it did, that generalization would be wrong, as studies conducted in the last decade show that conception is most likely to occur in the days leading up to ovulation, with the single highest statistical chance of conception being from having sex on the day preceding ovulation.)
Some people will tell you to count backwards 14 days from when you're expecting your next period, if you want to determine when you're ovulating (and when you're most fertile). That is also a generalization that may well not apply to a particular individual in that it assumes a 14 day luteal phase -- which is that portion of your cycle between when you ovulate and when your next period begins.
The easiest thing to do to start to try and get pregnant is to have sex every other day from, let's say, a week into your cycle.
If you want to get more involved...if you want to figure out when you are at *your* most fertile and/or maximize your chances of conception, you can use ovulation tests to determine when you're probably going to ovulate, or you can chart your cycles (which involves taking your temperature, and checking your cervical fluids and cervical position).
Sorry to go beyond your question, but if you're asking because you're trying to conceive, well, I thought you might find the additional information useful.

Everything is always done by the FIRST day of your last period. It also depends on how long your cycle is. For most people it is 28 days, so yes, 14 days after the first day of your last period. If you know your cycle, there are a lot of websites which will give you the dates if you punch in your information, like

Count from the first day of your cycle...period starts = day 1...then have sex from day 10 to day 16 every other day...for maximum chance of conceiving....you can also use ovulation calculators online...Good Luck!
