It means that you have surprised someone by how well you do something that you normally would not do; for example, if you work everyday in an office, but you take a dare and go up on stage and sing a song like Whitney Houston and surprise everyone with your talent, they could say, "You missed your calling," (as a singer)...essentially, that you're not using the great talent God gave you at that point.

"You missed your calling" means that you are not using the gifts God gave you and instead are choosing a different career path.

For example: I have a natural gift as an artist at all types of mediums and have an excellent eye for art, but I work as an administrative assistant.
