Who knows when the decision was made, but his verbal order to Goering in the Fall of 1941 initiated the mass killings. There's no written order to that effect, but it is known that Goering then ordered Reinhard Heydrich to begin the implementation. Heydrich convened a meeting of the top SS and Nazi bureaucrats in Jan. 1942 at the so-called Wansee Conference where the "Final Solution to the Jewish Question" was decided.

The Jewish people were educated, rich, and owned many businesses. The Nazis needed someone to blame the economic woes of post war Germany on. They had an ideal of a perfect Aryan race in there minds. The Jewish people did not fit the ideal and were the perfect scapegoat for the state of the economy. Jewish children grow up to be Jewish adults. The propaganda began to be spread and all Jews became the target of hatred, no matter what age. The old saying, "all it takes for evil to triumph is for a few good men to do nothing" became a reality. It was decided among his officers and himself, just before he took full control of Germany, to blame the Jews and not the Communist as was suggested.

The answer is not so simple as people believe it is.
Clearly, Hitler wanted to clear Europe of Jews and Gypsies. Several hundred thousand German Jews emigrated in the 30s, though -- like Castro's Cubans without a dime in their pockets. Many of those who emigrated into Western Europe had no idea that Hitler would soon conquer their lands of refuge.
Really, the first Nazi Judenfrei idea was to send all Jews to some place nobody cared about, like Madagascar. With the outbreak of war those things became impossible. Most German Jews had been driven out, but military success brought them many more, including German refugees.
The establishment of extermination camps began only in early 1942, and all were in conquered eastern territory. Poland and the Ukraine held many more Jews than Germany ever had, and the first idea was simply to kill as many as possible. Many were shot to death, but in a later reevaluation it was decided by people like Himmler that it was more efficient simply to establish death camps -- most known, Birkenau next to a known concentration camp, Auschwitz -- gas them to death and dispose of the corpses in ovens.
God, I'm getting sick even talking about this.
Anyhow, many of the living corpses discovered by US and UK troops in places like Buchenwald and Dachau had been marched west on no rations to avoid the advancing Soviets.

I dont think there was an exact date, though his stay in prison where he wrote Mein Kampf marked the first known hatred of Jews, who he blamed for Germany's problems.
