You could start by taking a canal trip. Embarkation sites are close to your hotel. Stroll down the Dam (main shopping street). Visite Mme Tusseaud museum.
Visit Rijksmuseum . Have a look at The Nightwatch by Rembrandt. Visit one of the diamond museums. Visit the Van Gogh museum. Visit the hippies in Vondel park and Leidse plein. And lastly visit the "walletjes" (red district) where you can admire the ladies of the night invery scant attire. But do not try to take pix or you could have very serious troubles.

All of the following are in the city and easily accessible. The #2 or #5 tram will get you almost anywhere you need to go. The canalbus is another cool way to get around, if you want a different perspective.
The Rijkmuseum - great collection of Rembrandts, including "Night Watch"
The Van Gogh Museum - if you are into Van Gogh and impressionism.
The Anne Frank House - very emotional experience, and I'm not an emotional person. Go early in morning or late afternoon, otherwise lines can be pretty long.
Vondelpark - Amsterdam's "Central Park" - great place for a stroll, picnic or leisurely bike ride
Also, be sure to check out one of the "brown cafes" - old establishments with walls that are literally brown from years and years of nicotine build-up. Nice places to relax and have a Heineken.
Of course, Amsterdam offers plenty of "coffee houses" if you are into that kind of thing. And while you can probably get a cup of coffee at one, that is not what most people go there for. If you decide to give it a try, I would make it your last stop for the day, and make sure you have your bearings and know how to get back to your hotel before you start "drinking coffee", as the tourists sometimes find the local "coffee" a little stronger than they thought. :)

If the weather is nice, it is always good to go to Museumplein during the day for people watching or some pick-up football.
The Leidseplein area is always interesting at night, with street performers and about any kind of food/nightclub/drink/hangout/live music you can think of. This area also has a very popular English language comedy club called Boom Chicago (be forewarned, it gets very smoky in there, but they may have a smoke-free show one or two nights).
I would also strongly suggest eating some Indonesian food while in town. The best (and possibly cheapest) place in town is Sari Citra on Ferdinand Bolstraat (a short walk from the Heineken brewery).
I would also highly suggest renting a bike (if the weather is good). Just go exploring away from the typical Dam Square touristy area and see what you find. The city proper is very small and every tram stop has a map just in case.
More than anything, just bring a good spirit and comfy walking shoes, and you'll be doin' fine!

Take the canal tour. It's very interesting. You could go to Corrie Tin Boom's place and also the Anne Frank house. There is also the Van Gogh museum and the flower auction. You can take a ride to the countryside which to me was awesome! Also, it's a short ride to the North Sea.

be careful in certain areas! there's a lot of drugs here and although some are legal, the way they're sold in Amsterdam isn't.haha
but you could visit historical areas, there is a lot of historical sights honoring the victims of world war two
