It depends on what caused the stain. Here's a chart to guide you:

I have found that the old fashioned methods work best for me. My main stain remover is a "bar of Sunlight Soap".
First wet the stained area, then lather it up with the bar of Sunlight soap, rub the area of the stain with another part of the polo shirt. Using a brush could possible ruin the area of the polo shirt that is stained, but if you rub it with the shirt itself you won't damage the weave.
I then apply another lather coat and let it soak in for about an hour or so. Finally, wash the shirt in your washing machine.
Before putting it into the dryer .... make sure that the stain is gone ... otherwise the heat in of the dryer may set the stain. If the stain is still there, repeat the process again and hopefully your polo shirt will be as good as new.
I find that this method works great on any kind of greasy stains. I hope it works for you .... best of luck!
