Hello.
This is my suggestion:
A)To go Lisboa by train because it is much more comfortable and quicker. You will pay more then going by coach but i think it is worth. You can check the train price and book it here. There is option for English (it's a tiny quarter of union jack flag)

B)What can you do there?
Well,it will also depend on your time but here are some tips.
Visit Castelo de S.Jorge (Saint George Castle) in Martim Moniz. From there you can see the city (down town),the river. Very beautiful view!
You can walk up there (good luck!)or you can go by mini bus or by tram. I think is more fun if you go by tram. When you go there (to the castle), on your way you will have the opportunity to see the old Lisbon:the narrow roads, allies, other passage which is just stairs and stairs ,the typical Portuguese ladies…it is just so traditional! Just love it!
After that you can walk to Rossio and Praca da Figueira (Figueira square) to do some shopping like clothes, shoes, souvenirs or just for a coffee. There are also very good restaurants.
Others things to do near by: Elevador de Sta.Justa (Saint Justa Elevator) to see the view of city from up there…..and have an ice-cream.Lol. Elevador da Gloria (Gloria’s Elevetaor): now this is just amazing! It is a tram that goes up a hill. This will lead you to another traditional part of Lisbon: Bairro Alto: Cool if you go in the evening there are very nice cafes just to chill and relax.
You can go to Parque das Nacoes (where was held the Expo 98). Here you can visit the aquarium, there is also a shopping centre which is open till 11pm. Loads of bars and cafes by the river side to chill and relax!
Belem, Mosteiro de Jeronimos, Padrao de Descobrimentos are other things to see. Just get a tram from Praca da Figueira and enjoy the view along the river side. Don’t forget to try the famous Pasteis de nata (custard cake) de Belem!!!
I recommend you to visit these 2 websites:

Must Dos in Lisbon.
Visit the Castle: Awesome view!
Visit Parque das Nacões and visit the massive Aquarium + other exhibitions
Visit the Benfica stadium!
Visit Belém and make sure you eat a custard tart!
Visit Mosteiros dos Jerónimos and admire the architecture!
Visit Estoril and Cascais!
Visit Sintra!
Train is the civilised way to travel from the Algarve (and cheap compared to UK prices!)
For Parque das Nacões arrive at Gare do Oriente.
Boa viagem! Enjoy Lisbon!

Coach is cheaper but takes an insane amount of time. Its best value for money to get the train, even though it is a little more expensive it is still cheap in my opinion.
P.S. You will have to go to Faro to get the train
