Yeah you should let her know! Don't be rude about it or anything just tell her, " Hey, I know you probably didn't mean for it to happen but what you said really hurt my feelings" If she's any kind of a friend she'll understand and it won't happen again.
P.S. you may want to pull her off to the side and tell her it may go over better

are you sure? because sometimes even if you are acquaintances, girls tend to flirt by making fun of the ppl they like, esp from the ages of infancy up till 16 or 17. after that, they get more forward and complicated.
but if not, i think you should definitely let her know that you were put off by it. don't be mean about it, tho, or else all the friends in between you might have to choose who they'd rather hang out with.

I don't think there's anything wrong with letting her know she hit a nerve. If she did not mean it in a bad way then she will probably be sorry for hurting your feelings.
On the other hand, if she did say it to be mean... well then don't talk to the horrid creature again.

Yes, you should inform her that you don't like being teased. If you don't like something, make sure you tell the person so you don't have to put up with it.
