...ribbed combed cotton or spandex, then sew the two together would that make my original fabric more "stretchy"?

If your polyester is a woven fabric it will stretch a little bit if you cut it on the bias (if you have a square of fabric the bias is a 45 degree angle from the center of the piece or if you draw a mental line from one upper corner to the lower opposite corner). It will still not stretch as it would if it had spandex or if it were a knit but it will drape & move better on the bias.

No fabric you can put over the top of it will change the actual stretch of the fabric beneath.

Sewing on the bias can be tricky but w/some practice & patience you'll become a pro!

No. The bottom layer just wouldn't stretch anymore because it would be stuck to something that didn't. You will just have to deal with the material as it is.
