I have a computer with ASUS AI motherboard. I have one hard drive in SATA 1 but the hard drive with adapter is in slot 2 and the computer is not recognizing it.

Do you mean a IDE to SATA "power" adapter so that your regular power supply can connect to the SATA drive?

And I'm assuming that you have connected the SATA data cable from this drive to the motherboard in the SATA2 slot...right?

Is the BIOS recognising the 2nd drive during POST? If not, you may have to enable the SATA2 in your BIOS. Check to make sure all SATA connections are enabled. And don't select the RAID configuration unless that's what you intended the additional drive to be.

If the drive is being recognised during POST, then you may have to adjust Windows or install a driver for that SATA connection.

At any rate, you should consult the little booklet that came with your montherboard.

ADDED:
Gold Dog made me recall that if you have a motherboard that does not recognise the faster SATA drives, you will need to set the jumper on the drive to restrict it to the slower transfer speed that the motherboard recognises.

Make sure you put the jumpers on the correct pins on the back of the hard drives. Otherwise they may not be configured correctly for what you are trying to do, or they may even conflict with each other.
