Starch is basically the polymer of sugar. The leaf has been destarched so there is no starch present at the start of the experiment by placing the leaf in a sugar solution you provide the sugar which would normally be produced by photosynthesis. The plant leaf still contains the enzymes required to synthesise the starch and so the glucose molecules are used to produce starch.
The best sugar solution to use would be glucose because that is the monomer unit used to synthesise starch but you could use sucrose (the sugar you buy in shops for cooking) which contains glucose. If you wanted you could compare the rates of starch synthesis with different sugar types.

Thew most commonly used test for starch is iodine, which turns black on reaction with starch. You could take samples at different times and test for starch to see how quickly the starch is seen in the leaf.

Destarched just means the leaf has not photosynthesised for some time and the stored resources(i.e. starch) are utilised so that the leaf does not contain any starch. The leaf still contains chlorophyll and thus can photosynthesise to produce sugars, which are converted to starch for storage.

I don't understand why you have to place the leaves in sugar solutions.
