Restriction Fragment Length Polymorphism (RFLP)

DNA is cleaved, and analysis is preformed on the cleaved DNA.

1. Isolate your DNA from tissue, or bacterial cells. You may need to amplify the amount of DNA you have by PCR.
2. Cleave your DNA with Restriction endonucleases. These enzymes will cleave your DNA at particular sequences only, so fragment sizes will vary.
3. Run your DNA thru a agarose gel. This will separate the DNA out by fragment size, resulting in a banding pattern.
