When the thought of being with the person one more day becomes more painful than the thought of losing them, that is when you know you are ready to end it. If the thought of losing them is no longer painful, it's already over, you just haven't made it official.
Some relationships have just become boring, and you need to try SOMETHING new in your relationship, not SOMEONE new.
Only you can decide which it is for you.

I'll assume you are talking of a love relationship.
You know a relationship is over when you think of the person less often (forget to call, email, sms), when you remember things that happened/you used to do together as if it was centuries ago that it happened, when you have to think of what to say b4 making a call/writing a letter or email ( you need a reason to do it), when your phone conversations get shorter (you feel relief that the conversation is over), when you dunno what life would be like with the person closer to you, etc.

oh my stuff in the yard is a good clue.
Or when you say to yourself "This is the last time I am gonna get treated this way.
Or when you make plans to save up money and move out.
Finding your person with another is a good clue, also.
When you feel drained and tired around them.
When their friends are criminals, or problem people.
When you could either go on a date with them or go to a movie yourself, and couldn't care less which, yeah, that's a clue that it's over.

When your boyfriend goes to K-Mart and gets you a $5 stuffed animal on sale for your birthday. Or when your boyfriend deliberately tries to antagonize you into an argument so he can use it as an excuse to break up with you. Or when your boyfriend tells you that you are smothering him or he needs more space.
