I've done this drive myself so I can tell you of my own experience. (1) LA to SF will take approx. 7 hours or a bit less if you take I-5 instead of 101S. (2) SF to LV will take approx. 9 hours.
I did both of these drives during the middle of the night (2 or 3am) to avoid traffic and to drive a bit faster (80-90 miles/hour).
If there is a way for you to stop in SF first instead of LA, it would definitely be a much better plan for you. You actually have to travel towards LA from SF to go to Las Vegas; so if you can find a way to stop in SF first then travel to LA, and from LA to LV it would make much more sense and save you lots more time!

how many times do you get to get away let alone drive the California coast line?
Take a extra day for this drive don't be in a hurry because if you are fly
take hwy 1 all the way from Malibu to San Luis obispo, maybe stopping in pismo beach to see it. then before you hit, the Hearst castle load up on drinks food and gas because the best part is about to happen but it is expensive from here on up[,Ragged point inn is the first place you come to 14 miles after the Hearst castle a beautiful place to see the entrance to the greatest meeting of the land and the sea, where you will see at the very point of ragged point majestic mountains falling into the pacific ocean. 3 miles north look for some cars parked on the side of the road for there is no parking lot look into the canyon and you will see a spectacular waterfall called samon creek, you can hike right up to it and take pictures in front of it. 8 miles after that is a community of gora famous for whale watching but as i said expensive.14 miles is Lucia lodge but just before you hit it 13 miles you will see a small sign that says Immaculate heart hematite, comedian monks run this place and the public is allowed up the one mile drive way witch is one of the only paved driveways in big Sur once you get to the top the views are spectacular,at the bottom Lucia lodge was made when the Hwy 1 was built in 1932. nestled at the point is 4 cabins and 6 rooms No TV"s that views are the natural TVs, 15 miles north is a place created in the 60's during the rainbow festivals movement a place were Hollywood Moguls come to get away and take there minds into another place. The esalen Institute is for you mind and mind only. 800 dollars a weekend to go into a room and scream to relieve your tension and you get to bunk with other holistics plus the tofu dishes are well worth 800 dollars ( NOT), but it has helped many of people got to the gate when your see the sign that says bu reservations only and get the latest edition of the classes they teach here, it will make a g coffee table conversation book,These classes like open mind training, upledger viceral manipulation, and others are for your mind. if this place can't relieve your stress no place will, that is the south coast big sur the next are all on your own and they are well known find a copy of El SUR GRANDE and it will tell you the rest .

Easiest way to find out is to go to maps on Yahoo and look it up.
5 hours and 17 mins to get from LA to San Fran according to Yahoo Maps.
14 Hours and 16 mins to get from San Fran to Las Vegas.
Good Luck and have fun!

I'd concur with Mark-- if you're just visiting and have any time at all to kill, take Highway 1 along the coast, its arguably the most scenic drive on earth.
I-5 is the fastest route, but it's also boring.
Also SF-LA-LV would be a lot more efficient that LA-SF-LV.

What route will you take, what speed will you drive.
How often will you need to stop for fuel. Too many variables to give an exact answer.
