Basically it's all about practice and repetition. It doesn't have to be all on the court though. Start on the court, doing things like the basic layups and free throws because you should have that fundamentally down. Then just keep shooting jumpers and trey's with your weak hand. When you're not on the court, start answering the phone with your weak hand, drinking with your weak hand, and doing things like opening the door with your weakhand. These small task will get your mind thinking both ways, then you can move up as you get better. It took me about two weeks to master those things, then I started tieing my strong hand behind my back to force me only dribbling with my weakhand and protecting it against my teammates in practice and using a screwdriver with my weak hand and bouncing a tennis ball off the wall and just catching it with my weakhand. Things like that should help you so you eventually have no weak hand at all and increase your game

When I was young I started eating with my other hand and writing (it wasn't pretty) and it was more mental than anything. But when I started to translate it to basketball I started dribbling the basketball first with my weak hand and then shooting layups first and then shots close around the bucket (4 or 5 ft). Playing horse and making everyone shoot with their off hand also helps and in no time you should be able to shoot free throws with you off hand....

Favor your weak hand over your dominant. This will allow it to develop the muscle function and muscle memory that is needed to do certain things.
A good exercise is to stand below the rim, slightly to the side of your weaker arm. Use that hand to put up short shots off the backboard.
It really just boils down to practice.

Just practice a lot. I sucked with my weak hand before. I started practicing and now I can do almost as much with my weak hand as my dominant hand.

just plain practice.
In scrimmage, be sure to use your non-dominant hand to make layups on that side....it will come.
On a breakaway, practice switching side to side with the dribble.

like the first two answers said, it just takes practice. i am right handed and i really practiced with my left hand, dribbling, shoot layups, hooks, etc.
i finally got to where i favored dribbling with my left hand to set up my right handed jump shot.
