when using a micrometer, it should be calibrated for you, and if not, it can be done using a commercially prepared slide. in this way, you can calculate what each increment on the scale is in regards to microns. this is calculated for each objective. then just use the micrometer, count how many units long the cell is, and simply multiply by the factor calculated for each objective. this has to be done for each individual scope in use, as it can vary from one to another.
