It depends on what you are looking for.
Since I just got back from Australia and had a trip to the Caribbean two years ago, I know I would not mind going back to either place... as well, I had an old girlfriend who visited Spain and fell in love with it, so I imagine it is pretty good too.
I loved Australia - friendly people, unique things to see and do, lots of different wild life and "wild life". It is a huge country that would take several weeks to see. If you are looking for an activity-filled time, Australia is a great place to visit.
The Caribbean is more laid back and relaxing. Again nice people, but somewhat more commercialized and filled with tourist traps.
As for Spain, I would expect it to be a different cultural experience that the other two.
All three have beaches, sun and great people. If that is all you are looking for, you can't really go wrong with any of them. However, if I had to pick only one, it would be Australia.

vote caribbean (as long as I can island hop).
Have travelled quite extensively round the other two and would happily go back, but have only bene to one place, Antigua, in the Caribbean. If I hadn't been to Aus and/or Spain, I would have chosen a destination that I haven't been to.
