Possibly. In the eukaryote, genes occur on both sides of a chromosome, or on each pair. Therefore, if it is a dominant trait, both alleles will need to be knocked out it order to gain a mutant phenotype.

Not really, selective breeding will accomplish this but it is doable with only one vector and exteme selection techniques in ES cells.
