The first thing you do is learn the hand rankings, these are common to all games that don't have a wild card or a low pot (most of them).
Once you have learned the poker hand rankings you need to decide which game you want to learn. The most popular game right now is Texas Hold'em. In this variation of poker you get two private cards for you, and five cards are dealt face up in the middle of the table. You have to make the best five card poker hand using three, four or five of the community cards, together with your cards. So you might use one of your cards, with four of the community cards, or both your cards with three community cards. Sometimes the community cards are best without either of your cards.
For more details follow the link and check out the beginners section. Consider buying Poker For Dummies (you don't have to be a dummy, it's a great book) or some other beginners book to learn the game. Then play for free at one of the poker sites using play money. This is easy and fun, since the computer prompts you when it is your turn, and lets you know what you are able to do. You should be able to learn the game in a few hours. But the bad news is to become really good at the game takes years of practice for most people. But it's a fun, interesting and rewarding journey.

There's way too many forms of poker to explain it in one post. A site like http://www.theultimatepokerguide.com has the rules for all of the forms of poker which will explain it a lot better than I could in any answer. The games are all so different from each other that if you post any general answer ti will never be accurate for all of them.

These are all rules specific to the game Texas Hold 'Em, which is a variation on poker. "Poker" in and of itself can represent many different games, such as Hold 'em, 5 card draw, 7 card stud, Omaha, etc. Hold 'em is the most popular today, but 5 card draw is probably the simplest and more popluar among beginners.
