With no hold ups you can do it in under 3 and a half hours by taking the A660 out of Leeds, joining the A65 just beyond Otley, follow the A65 to the M6 then sraight up the motorway to Glasgow, watch the road signs carefully in Glasgow, if you keep going straight on you'll end up heading out of Glasgow towards Cumbernauld and Stirling.
Avoid the A65 at busy times and definitely when there's holiday traffic - it's a single track road most of the way and can be very slow going when it's busy. The alternative is the M62 towards Manchester then the M61 to join up with the M6, again, watch the road signs or you'll miss the M61. Journey time this way is about 4 and a half hours.
In the rush hour avoid the M62, the stretch between junctions 27 (M62 / M621) and 23 (Ainley Top, Huddersfield) can slow to an almost standstill. Take the M1 north then the A1 to Scotch Corner then the A66 across country to the M6 at Penrith. This way will take you about 5 hours.

It takes around 4 hours. Drove down last year in the dark, would recommend going by daylight and take a snack. thought it would take 3 hours. we went M74, via scots corner way rather than manchester way which friends in leeds who lived in glasgow said was the way to go
