Both spellings I think are nice, but I would go with Anna. First of all, I would pronounce Ana like ah-nah or aw-nuh, so I wouldn't want it mispronounced. Also, for spellings reasons. My name is Claire, and I get extremely irritated when people spell it Clair or Clare, and I can guarantee you that people will spell Ana like Anna ALL the time. Oh, and if you ARE pronouncing it Ah-nah, then definitely spell it Ana.
Middle namews for Anna:
Anna Beatrice
Anna Bethany
Anna Brielle
Anna Catherine
Anna Claire
Anna Danielle
Anna Elizabeth
Anna Gabriella
Anna Isabelle
Anna Kirsten
Anna Kristen
Anna Josephine
Anna June
Anna Laurel
Anna Lauren
Anna Margaret
Anna Madelyn/Madeleine
Anna Olivia
My favorites are Anna Beatrice and Anna Josephine, good luck deciding =)

I like Ana better, Anna looks a bit old fashioned as if it were Anne, which is a pretty name, but decide whether you would want it more modern or not. I prefer modern. And for the middle name, maybe Jenelle or Serene? I think they are both very pretty names. This is what some might sound like:
Ana Jenelle
Ana Serene
Anna Christine
Anna Jacquelyn
Maybe you could honor someone in your family who has passed away or in their last years with the middle name? Anyways, congratulations!! And if you decide not to name her Ana or Anna, here are some other suggestions:
Jacquelyn
Zoey
Zhanna
Jenelle
Christine or Christina
Hedley
Brooklyn
Ashlynn
Amber
Maya
Serene
Gabby
Gabrielle
Serenity
I hope I helped! Good luck and congrats!!

The thing is, with Ana, people might say (AHN-uh) instead of (ANN-uh). Anna or Annah is better, I would say.
As for middle names...
>Anna Rosemarie
>Anna Arian
>Anna Azul
>Anna Antoinette
>Anna Ariella
>Anna Bernadette
>Anna Delani
>Anna Elaine
>Anna Felicity
>Anna Gabrielle
>Anna Nicollete
>Anna Juliet
>Anna Leilani
>Anna Lilianne
>Anna Mariel
Also, check out

and go to the "3+ Syllable names that don't end in A, and are not stressed on the first syllable (the Next Generation's Elizabeths) " section or the "1-Syllable Names (the Anns, Waynes and Lynns of Tomorrow)" section.

Well, I know quite a few Anna's, but not many Ana's. I'd go with Ana... Not as common. (sounds odd when read aloud. haha)
As for middle names, I like Ana Rose, but that depends on the last name.

I would go with Anna. its the traditional way of spelling it. If I saw the name Ana I wouldnt know how to pronounce it. I would say it more like ON-A. I also agree wit hthe person who said that it will get misspelled alot and she'll always be correcting people. or you could do what my husband and I plan to do. he loves the name Anna but I dont like it too much so we are going to name our daughter Aubrianna so he can still call her Anna for short, but its not actually her first name.

Ana is Ah-na Anna is An-na so however you intend on pronouncing it is how you should spell it. I like Anna Juliet or Ana Michelle. Other options are using as a nickname for something like Juliana or Mariana; my niece is Cynthiana even Anastacia or Anabelle would work well. Good luck and have fun choosing!
