Different slot types in the computer. AGP cards only fit AGP slots. PCI cards only fit in PCI slots. AGP is preferred over PCI.

It refers to the type of slot in the motherboard the card will fit into. Motherboards should have so many PCI slots, so many AGP slots, etc., so you should try and find out what slots your motherboard has before going out and purchasing a new card so you don't come back and find out there's no where for it to go!

PCI-Express are the newest kind and most new cards are made in this format (which is different from standard PCI, by the way).

Well AGP is better than PCI, but PCI Express is the best, it is different than PCI. See below for detailed explanation of PCI Express.

PCI - 1st Generation
AGP - 2nd Generation
PCI Express - 3rd Generation

PCI Express (Official abbreviation PCIe, PCI-E often used, not to be mistaken for PCI-X) is a computer system bus that allows expansion cards with various capabilities to be added to a system. It is a flexible system intended to replace PCI, PCI-X and AGP. While PCI Express has the same software interface as PCI and can be bridged to PCI, the cards are physically and electrically incompatible. While in development, PCI Express was referred to as Arapaho or 3GIO for 3rd Generation I/O.

PCI-E (PCI Express) is the newest type. AGP is better than PCI, but PCI-E is better than AGP. If you have a newer computer, you probably have PCI-E ports.
