I presume that you have created a sub-domain through cpanel or some other means with your server. The subdomain should look like this: http://subdomain.maindomain.com.
Now I also presume that you are using FTP to upload files. So when you open your ftp, the first thing you should see is your main domain.
Click on the public_html folder of your main domain and you should see, amongst other things, a folder with your subdomain name on it. Click on this folder to open the public_html folder of your subdomain. Now upload your index.htm (index.html) into this folder.

Blimey Rex, you make that sound complicated. ;o)

It depends on the control you have over your domain
name, and the server it points to. Basically a sub-domain
should be considered as a domain name in its own right,
entirely separate from the original name it is based on.

Unless you properly control the domain name ( let us
assume in this case that you do ), then you can't create a
sub-domain.

A correct DNS entry will resolve your sub-domain.

A correct and entirely separate entry in the web server
configuration file will allocate space for your new
domain name ( sub-domain ).

Should you not have this kind of control, then it is also
possible to fake a redirection.

HTH ( a little ).

If both the domain and the subdomain point to the same physical server, then your server software (Apache, IIS, etc) must be configured to handle the HTTP header in the request and determine whether it is for a page in the domain or the subdomain.
