This first bit may be obvious but choose a subject you like, as it has to be humorous pick something you enjoy doing.

Firstly you will know something about it and wont need to do to much extra research, you are also likely to have had some 'funny' experiences or seen something 'funny' happen to someone else.

So if you picked riding a bike.

You could do your speech from either 'How to..' or 'How not to..'

In terms of persuasiveness in this area you would be aiming your talk toward people who have never tried to ride a bike so, how much fun you find it, it gets you places more quickly and other positive reasons for doing this activity.

I am assuming you know how to structure a speech, but a last tip.

Don't write it long hand and read it off the paper or 'memorise' it if you do it will sound boring and automated.

The key to any good speech is retaining the audiences interest so use 'one word reminders' on a piece of paper in case you lose your thread, hold your head up look at your audience and project your voice to the back, move your vision around the audience during your speech so you indicate you are interested in all the people in the room.
Also this conveys confidence that you are comfortable in speaking in public. It impresses people and they will be more attentive to what you say.

The best thing to write a speech about is something that you know a lot about - if you have any knowledge in an area you will speak more from the heart and less from the paper - you'll get your point across more effiecently.
Could you use a favourite TV show as a starting point? Take a current issue from a Soap?

Mike the Headless Chicken

I did one before, and the audience died laughing. It was really interesting too. Pick something that interests you, but something that you won't laugh at while giving the speech.
