It really depends highly upon what toner cartridge you are using.
It is not advisable however for the following reasons.

1. Toner dust is highly carcinogenic and should not be inhaled. It is also a very find power which makes it easily airborne.

2. Toner cartridges are often smart shipped, without a replacement chip the toner will not function.

3. toners have several moving parts that do have a service life. If these parts breakdown you run the risk of ruining your printer.

Yes, but you'll probably need to buy a kit. Many laser printers have a chip that keeps track of the level of toner in the cartridge so the printer won't work right if you refill it instead of buying a new one; a good kit will contain both more toner and a replacement chip so your printer keeps working. Did a refill myself last year; it's not too difficult, but be sure to put down newspaper as it can be messy.
