When my microphone wouldn't work, I eventually found that it was set to use a front panel jack - even though that wasn't actually connected!

right click on the "volume" icon in the taskbar
select "Open volume control"
options, properties - check "Microphone" - OK
options, advanced controls...
click on the microphone's advanced button
make sure that 'Front panel microphone' is unchecked. Close

If you removed the add in PCI sound card, your BIOS should have reset to read the onboard.
Do you get sound out of your speakers? I'll assume yes for now. Chances are its a Realtek AC97 onboard sound, so go into your control panel, open up the sounds and devices and make sure the thing isn't muted. May sound lame, but you'd be surprised.
If your not getting any sound, go into the BIOS and enable the onboard sound.
Always fund starting at the beginning with the little things worked the best.
Good luck to you. It's probably something really silly.

Try the simple things first. We assume your mic is working properly. Make sure that in the volume control panel that the mic button isn't muted. Sometimes it mysteriously gets checked.

If that doesn't work, go into control panel uninstall any software programs and then system and uninstall both the on-board and add on card. Reboot, Windows will find the new software and install the drivers for it. Usually.

Hope it helps.
