I am thinking of creating a business directory site as a business, but I don't know how it can generate money to cover the expenses involved and to make me run the business at a profit. Anybody with an idea how this type of business works should please enlighten me.

Put Adsense ads on it and then register it on the web everywhere you can. The traffic should pay for the site.

Advertising and/or memberships. That's what makes the Internet information world spin. :)

Banner ads (generally) can be either text based (like with Google AdSense, Yahoo Publisher Network, Adbrite) or remnant inventory based (like with advertising.com, etc) OR through direct sales. There's a million ways to get into the advertising world online.

Memberships are clear and simple. Offer a service, charge for it.
