i dunno, it depends on your taste. if you are a die-hard action/epic fan then 300 but if you like your movie to have some substance+great acting+even better direction then The departed is made for you.
personnaly i prefer The Departed but damn you for making me choose as they are both awsome films in their own rights.

"300" is junk food. "The Departed" is a steak. It depends upon your mood (and probably age). "300" is not in the same league as "The Departed" (Best Picture).

The Departed had a better script, cast, and direction.
300 had better visual effects and sound.
They're in two different categories, it's like comparing Jurassic Park to Citizen Kane.

300, Departed was really hard to get into and it seemed Martin really enjoyed his blood sprays
