The best one out there is the Nissan Altima-it gets 42 miles per gallon in the city, which I believe leads all non-hybrid sedans. It does, however, start at $24,400. If this is outside of your budget, I would say go with the Honda Civic-It doesn't get 40 miles per gallon, it gets 38, but it only starts at $15,010. The only two non-hybrid cars out there that get over 40 miles per gallon are the Nissan Altima and the Toyota Camry, which gets 40 in the city, but it starts at $25,900, which is WAY too much money for that car. I strongly suggest avoiding the hybrids because in order to get high miles per gallon, you have to drive under like 25 miles per hour or else the car will just switch over to gas for fuel and no electricity and act just like a normal car...that's my belief, anyway. I think the Altima is a much better car than the Camry, anyway. Check out this link for other cars that get high miles per gallon: http://www.automotive.com/fuel-efficient-cars/109/sedans/30-mpg/index.html
The Ford Fusion is also a cool new all wheel drive car that gets good mileage: 24 combined city and highway. It also starts at a very reasonable $17,995. I really like this car and I think it will be a real winner for anyone who buys it. My belief is that it will last as long as any other car out there. Keep in mind it does have all wheel drive, which will lower miles per gallon a bit, but I think that all wheel drive is an extremely handy feature to have in any car.
If you're more of a luxury car kind of guy, then the best option would be either an Audi A4 or A6....they don't get anywhere near 40 mpg, though...they get around 26 or 27. Again, they both have all wheel drive-the Audi Quattro all wheel drive system, which is the best all wheel drive system in the world, and again, all wheel drive is an extremely handy feature and I would recommend it in any car, even though it may drop your miles per gallon a bit. The BMW 5 series also gets decent miles per gallon but it is in the same price range as the Audi but doesn't come with all wheel drive. BMW's are also extremely expensive to fix. I have driven both; believe me, the Audi is MUCH better in every category. The handling of Audis is exquisite!
By the way-in case you were wondering, the major hybrid out there, the Toyota Prius, gets 51 estimated miles per gallon in the city and an estimated 60 on the highway. If this interests you, then go for it. All of the cars that I listed above are normal gasoline-powered cars...none of them are hybrids because as I said before, I don't like the idea of the hybrid YET, but I think eventually all the kinks will be worked out and practically everyone will drive a hybrid. Hope this helps, and good luck finding a new car!

Short answer: the Toyota Prius.

Long answer: No cars made in the past few years beat 30 MPG by any distance, except for hybrids and diesels. Geo Metros and Ford Aspires hit 40 MPG, but they haven't been made in several years.

Some of the hybrids get 40 MPG... the Toyota Prius is a 4-door sedan or hatchback. The Honda Insight is a 2-door. The Lexus hybrids are large and classy but don't hit your MPG number.

High-performance diesels are coming later this year, thanks to a radical change in the diesel smog laws. Now that they've finally set smog standards for diesels, many new manufacturers are jumping into the diesel car biz, and I expect some of them will hit 40 MPG. The older VW diesels, Rabbit, Golf or Beetle get easily 40+ MPG, but they're awfully small. The Mercedes diesels don't quite get 40 MPG but people love 'em. All can run biodiesel and some can be converted to run straight vegetable oil.
